SIC offers sample handling options beyond on-line derivatization. By adding peripheral devices (pumps / valves / reaction chambers), even complex sample preparation routines can be carried out in an automated fashion. An example is on-line microdialysis, which can be utilized to remove protein contaminants from the sample before it is subjected to derivatization, separation and detection in the monitoring of bacterial culture (A) carried out by an SIC instrument (B). The details of this method developed by Lahdesmaki (2007) are summarized in the enclosed report.
Two overlaid SIC chromatograms obtained by monitoring cell culture fluid for 30 hours during which 27 injections performed in automated and unattended operation document the exceptional robustness and time stability od SIC technique. (Note: pump refill periods have been removed for this graphical representation).
